What Is The Difference Between Data And Instruction When They In Memory?

No difference from the content and physical point of view. The difference is in the moment when the memory where is stored is read. If it is read in the fetch phase, then it is an instruction, else it is a data.
This means that the same physical memory cell can be read as data and as instruction, if it is read  during the fetch phase, or as an instruction if it is read during the execute phase.
